Regulatory T-Cell Response to Low-Dose Interleukin-2 in Ischemic Heart Disease
نویسندگان
چکیده
This phase 1b/2a, randomized, double-blind, placebo-controlled, dose-escalation trial tested low-dose subcutaneous aldesleukin (recombinant IL-2) in patients with ischemic heart disease. Low-dose IL-2 expanded Tregs, without adverse events of major concern. Single-cell RNA-sequencing circulating immune cells was used to provide mechanistic assessment the treatment’s effects.

Low-dose interleukin-2 fosters a dose-dependent regulatory T cell tuned milieu in T1D patients.
Most autoimmune diseases (AID) are linked to an imbalance between autoreactive effector T cells (Teffs) and regulatory T cells (Tregs). While blocking Teffs with immunosuppression has long been the only therapeutic option, activating/expanding Tregs may achieve the same objective without the toxicity of immunosuppression. We showed that low-dose interleukin-2 (ld-IL-2) safely expands/activates ...
متن کاملEffects of low-dose recombinant interleukin 2 to promote T-regulatory cells in alopecia areata.
IMPORTANCE An impaired inhibitory function of circulating CD4+CD25+ regulatory T (Treg) cells was reported to play a key role in alopecia areata (AA). We report the first use to our knowledge of low-dose interleukin 2 for treating severe AA by promoting the recruitment of Treg cells. OBSERVATIONS We conducted a prospective open pilot study in 5 patients with severe AA resistant to previous sy...
متن کاملeffects of submaximal aerobic exercise on regulatory t cell markers of male patients suffering from ischemic heart disease
there are confirmed beneficiary effects of exercise on atherosclerotic inflammation of ischemia-associated heart diseases. the purpose of this study was to evaluate the effect of aerobic exercise on t-regulatory cell markers of il-35 as well as foxp3 and t-helper2 marker of il-33 in patients with ischemic heart disease (ihd). this research was performed on 44 asymptomatic male patients with isc...
متن کاملInterleukin-2 in relation to T cell subpopulations in rheumatic heart disease.
Interleukin-2 (IL-2) and T cell subpopulations were evaluated in children with rheumatic heart disease (RHD). Three groups were included: 13 patients with active RHD, 12 with non-active RHD, and 14 control children. Serum IL-2 and T cell subpopulations were measured by radioimmunoassay and monoclonal antibodies respectively. Patients with active RHD showed a significant increase in IL-2 concent...
متن کاملFailed adoptive immunotherapy with tumor-specific T cells: reversal with low-dose interleukin 15 but not low-dose interleukin 2.
Adoptive immunotherapy with tumor-specific T cells has emerged as a valid approach for prevention or treatment of diseases, such as melanoma and EBV-associated lymphoma. As interleukin (IL) 15 promotes survival of CD8(+) memory CTLs, we hypothesized that it could be used to enhance antitumor immunity in vivo through the maintenance of adoptively transferred memory CTL. To test this, we treated ...
